Although we have sent humans into space for more than 50 years crucial questions regarding kidney physiology, volume regulation and osmoregulation remain unanswered. The complex interactions between the renin-angiotensin-aldosterone system, the sympathetic nervous system, osmoregulatory responses, glomerular function, tubular function, and environmental factors such as sodium and water intake, motion sickness and ambient temperature make it difficult to establish the exact effect of microgravity and the subsequent fluid shifts and muscle mass loss on these parameters. Unfortunately, not all responses to actual microgravity can be reproduced with head-down tilt bed rest studies, which complicates research on Earth. Better understanding of the effects of microgravity on kidney function, volume regulation and osmoregulation are needed with the advent of long-term deep space missions and planetary surface explorations during which orthostatic intolerance complaints or kidney stone formation can be life-threatening for astronauts. Galactic cosmic radiation may be a new threat to kidney function. In this review, we summarise and highlight the current understandings of the effects of microgravity on kidney function, volume regulation and osmoregulation and discuss knowledge gaps that future studies should address.

Background: Pain is a very important factor in patient recovery and satisfaction. Following incision and drainage of perianal abscess, wound packing has been found to be very painful. The aim of this study is to compare packing the wound and not packing the wound after incision and drainage (I&D) of perianal abscess, taking into account recurrence rate, rate of occurrence of perianal fistula as well as post-operative pain. Materials and Method: Standard medical electronic databases were searched with the help of a local librarian and relevant published randomised controlled trials (RCT) were shortlisted according to the inclusion criteria. The summated outcome of post-operative pain score, recurrence rate, and rate of occurrence of perianal fistula was evaluated using the principles of meta-analysis on RevMan 5 statistical software. Result: Three RCTs on 490 patients undergoing I&D of perianal abscess were found suitable for this meta-analysis. In the random effects model analysis, the post operative pain score was statistically lower in NPG [risk ratio 0.66, 95%, CI (0.36, 0.97), Z=4.25, P=0.0001]. There was no heterogeneity [Tau2=0.15; chi2=9.32; df=6; I2=36 %; p=0.16] between the studies, however, statistically it was not significant. In addition, there was no statistical difference between the 2 groups in terms of recurrence of perianal abscess or the occurrence of perianal fistula [risk ratio 0.64, 95%, CI (0.31, 1.31), Z=1.23, P=0.22], [risk ratio 1.41, 95%, CI (0.89, 2.23), Z=1.47, P=0.14] respectively. Conclusion: Not packing the wound post I&D of perianal abscess is associated with significantly reduced VAS pain scores, which may result in improved recovery and patient satisfaction and has no effect on recurrence rate or the occurrence of perianal fistula. However, more RCTs recruiting a greater number of patients are required to endorse these findings.

Researchers point to four potential issues related to the popularisation of quantum science and technology. These include a lack of explaining underlying quantum concepts of quantum 2.0 technology, framing quantum science and technology as spooky and enigmatic, framing quantum technology narrowly in terms of public good and having a strong focus on quantum computing. To date, no research has yet assessed whether these potential issues are actually present in popular communication about quantum science. In this content analysis, we have examined the presence of these potential issues in 501 TEDx talks with quantum science and technology content. Results show that while most experts (70%) explained at least one underlying quantum concept (superposition, entanglement or contextuality) of quantum 2.0 technology, only 28% of the non-experts did so. Secondly, the spooky/enigmatic frame was present in about a quarter of the talks. Thirdly, a narrow public good frame was found, predominantly by highlighting the benefits of quantum science and technology (found in over 6 times more talks than risks). Finally, the main focus was on quantum computing at the expense of other quantum technologies. In conclusion, the proposed frames are indeed found in TEDx talks, there is indeed a focus on quantum computing, but at least experts explain underlying quantum concepts often., 30 pages, 7 figures

We introduce a fast solver for the phase field crystal (PFC) and functionalized Cahn-Hilliard (FCH) equations with periodic boundary conditions on a rectangular domain that features the preconditioned Nesterov accelerated gradient descent (PAGD) method. We discretize these problems with a Fourier collocation method in space, and employ various second-order schemes in time. We observe a significant speedup with this solver when compared to the preconditioned gradient descent (PGD) method. With the PAGD solver, fully implicit, second-order-in-time schemes are not only feasible to solve the PFC and FCH equations, but also do so more efficiently than some semi-implicit schemes in some cases where accuracy issues are taken into account. Benchmark computations of five different schemes for the PFC and FCH equations are conducted and the results indicate that, for the FCH experiments, the fully implicit schemes (midpoint rule and BDF2 equipped with the PAGD as a nonlinear time marching solver) perform better than their IMEX versions in terms of computational cost needed to achieve a certain precision. For the PFC, the results are not as conclusive as in the FCH experiments, which, we believe, is due to the fact that the nonlinearity in the PFC is milder nature compared to the FCH equation. We also discuss some practical matters in applying the PAGD. We introduce an averaged Newton preconditioner and a sweeping-friction strategy as heuristic ways to choose good preconditioner parameters. The sweeping-friction strategy exhibits almost as good a performance as the case of the best manually tuned parameters.

A new and efficient neural-network and finite-difference hybrid method is developed for solving Poisson equation in a regular domain with jump discontinuities on embedded irregular interfaces. Since the solution has low regularity across the interface, when applying finite difference discretization to this problem, an additional treatment accounting for the jump discontinuities must be employed. Here, we aim to elevate such an extra effort to ease our implementation by machine learning methodology. The key idea is to decompose the solution into singular and regular parts. The neural network learning machinery incorporating the given jump conditions finds the singular solution, while the standard five-point Laplacian discretization is used to obtain the regular solution with associated boundary conditions. Regardless of the interface geometry, these two tasks only require supervised learning for function approximation and a fast direct solver for Poisson equation, making the hybrid method easy to implement and efficient. The two- and three-dimensional numerical results show that the present hybrid method preserves second-order accuracy for the solution and its derivatives, and it is comparable with the traditional immersed interface method in the literature. As an application, we solve the Stokes equations with singular forces to demonstrate the robustness of the present method.

We present the development of a non-reflecting boundary condition, based on the Local One-Dimensional Inviscid (LODI) approach, for Lattice Boltzmann Models working with multi-speed stencils. We test and evaluate the LODI implementation with numerical benchmarks, showing significant accuracy gains with respect to the results produced by a simple zero-gradient condition. We also implement a simplified approach, which allows handling the unknown distribution functions spanning several layers of nodes in a unified way, still preserving a comparable level of accuracy with respect to the standard formulation.

Physiological solvent flows surround biological structures triggering therein collective motions. Notable examples are virus/host-cell interactions and solvent-mediated allosteric regulation. The present work describes a multiscale approach joining the Lattice Boltzmann fluid dynamics (for solvent flows) with the all-atom atomistic molecular dynamics (for proteins) to model functional interactions between flows and molecules. We present, as an applicative scenario, the study of the SARS-CoV-2 virus spike glycoprotein protein interacting with the surrounding solvent, modeled as a mesoscopic fluid. The equilibrium properties of the wild-type spike and of the Alpha variant in implicit solvent are described by suitable observables. The mesoscopic solvent description is critically compared to the all-atom solvent model, to quantify the advantages and limitations of the mesoscopic fluid description., Comment: 15 pages, 6 figures

Mandelic acid is an enantiomer of interest in many areas, in particular for the pharmaceutical industry. One of the approaches to produce enantiopure mandelic acid is through crystallization from an aqueous solution. We propose in this study a numerical tool based on lattice Boltzmann simulations to model crystallization dynamics of (S)-mandelic acid. The solver is first validated against experimental data. It is then used to perform parametric studies concerning the effects of important parameters such as supersaturation and seed size on the growth rate. It is finally extended to investigate the impact of forced convection on the crystal habits. Based on there parametric studies, a modification of the reactor geometry is proposed that should reduce the observed deviations from symmetrical growth with a five-fold habit.

We investigate the sedimentation dynamics of a binary mixture, the species of which differ by their Stokes coefficients but are identical otherwise. We analyze the sedimentation dynamics and the morphology of the final deposits using Brownian dynamics simulations for mixtures with a range of sedimentation velocities of both species. In addition, we use the lattice Boltzmann method to study hydrodynamic effects. We found a threshold in the difference of the sedimentation velocities above which the species in the final deposit are segregated. The degree of segregation increases with the difference in the Stokes coefficients or the sedimentation velocities above the threshold. We propose a simple analytical model that captures the main features of the simulated deposits.

We propose a multiple relaxation time entropic realization of a two-phase flow lattice Boltzmann model we introduced in earlier works arXiv:2112.01975 S.A. Hosseini, B. Dorschner, and I. V. Karlin, arXiv preprint, arXiv:2112.01975 (2021). While the original model with a single relaxation time allows us to reach large density ratios, it is limited in terms of stability with respect to non-dimensional viscosity and Courant--Friedrichs--Lewy number. Here we show that the entropic multiple relaxation time model extends the stability limits of the model significantly, which allows us to reach larger Reynolds numbers for a given grid resolution. The thermodynamic properties of the solver, using the Peng--Robinson equation of state, are studied first using simple configurations. Co-existence densities and temperature scaling of both the interface thickness and the surface tension are shown to agree well with theory. The model is then used to simulate the impact of a drop onto a thin liquid film with density and viscosity ratios matching those of water and air both in 2-D and 3-D. The results are in very good agreement with theoretically predicted scaling laws and experimental data.

The solution of Poisson-like equations defined on complex geometry is required for gyrokinetic simulations, which are important for the modelling of plasma turbulence in nuclear fusion devices such as the ITER tokamak. In this paper, we compare three solvers capable of solving this problem, in terms of the accuracy of the solution, and their computational efficiency. The first, the Spline FEM solver, uses C 1 polar splines to construct a finite elements method which solves the equation on curvilinear coordinates. The resulting linear system is solved using a conjugate gradient method. The second, the GmgPolar solver, uses a symmetric finite differences method to discretise the differential equation. The resulting linear system is solved using a tailored geometric multigrid scheme, with a combination of zebra circle and radial line smoothers, together with an implicit extrapolation scheme. The third, the Embedded Boundary solver, uses a finite volumes method on Cartesian coordinates with an embedded boundary scheme. The resulting linear system is solved using a multigrid scheme. The Spline FEM solver is shown to be the most accurate. The GmgPolar solver is shown to use the least memory. The Embedded Boundary solver is shown to be the fastest in most cases. All three solvers are shown to be capable of solving the equation on a realistic non-analytical geometry. The Embedded Boundary solver is additionally used to attempt to solve an X-point geometry, highlighting the problems with concave boundaries.

Liouville's equation describes light propagation through an optical system. It governs the evolution of an energy distribution on phase space. This energy distribution is discontinuous across optical interfaces. Curved optical interfaces manifest themselves as moving boundaries on phase space. In this paper, an ADER discontinuous Galerkin (DG) method on a moving mesh is applied to solve Liouville's equation. In the ADER approach a temporal Taylor series is computed by replacing temporal derivatives with spatial derivatives using the Cauchy-Kovalewski procedure. The result is a fully discrete explicit scheme of arbitrary high order of accuracy. A moving mesh is not sufficient to be able to solve Liouville's equation numerically for the optical systems considered in this article. To that end, we combine the scheme with a new method we refer to as sub-cell interface method. When dealing with optical interfaces in phase space, non-local boundary conditions arise. These are incorporated in the DG method in an energy-preserving manner. Numerical experiments validate energy-preservation up to machine precision and show the high order of accuracy. Furthermore, the DG method is compared to quasi-Monte Carlo ray tracing for two examples showing that the DG method yields better accuracy in the same amount of computational time.

S-ryhmän luontojalanjälki -hanke käynnistyi vuoden 2022 alussa ja tässä väliraportissa esitellään ensimmäisiä välituloksia. S-ryhmän arvoketjun ja oman toiminnan luontojalanjälki on laskettu käyttäen Jyväskylän yliopiston resurssiviisausyhteisön, JYU.Wisdomin, kehittämää laskentamenetelmää. Väliraportissa kuvataan laskentamenetelmän perusteet. Tulokset on raportoitu erikseen päivittäistavarakaupalle, käyttötavarakaupalle, tavaratalokaupalle, polttoaineille ja matkailu- ja ravintola-alalle vuodelta 2020 sekä organisaation omalle toiminnalle vuodelta 2021. Luontojalanjäljen mittarina käytetään osuutta lajeista, jotka ovat riskissä kuolla sukupuuttoon globaalisti (potentially disappeared fraction of species, PDF). Luontojalanjäljen laskenta osoitti, että S-ryhmä lisää globaalia sukupuuttoriskiä 0,0027 prosentille maaekosysteemien lajeista, 0,00037 prosentille makean veden ekosysteemien lajeista ja 0,011 prosentille meriekosysteemien lajeista. Arvoketju muodosti 94 prosenttia maaekosysteemeihin, 92 prosenttia makean veden ekosysteemeihin ja yli 99 prosenttia meriekosysteemeihin kohdistuvasta luontojalanjäljestä. S-ryhmän oma toiminta muodosti 6 prosenttia maaekosysteemeihin, 8 prosenttia makean veden ekosysteemeihin ja alle 1 prosentin meriekosysteemeihin kohdistuvasta luontojalanjäljestä. Yleisesti elintarvikkeet ja polttoaineet muodostivat suurimmat luontojalanjäljet. Laskentamallin mukaan globaalista luontojalanjäljestä alle 10 prosenttia kohdistui Suomeen, eli yli 90 prosenttia luontojalanjäljestä kohdistui Suomen ulkopuolelle. Toisaalta luontohaittaa aiheuttavista suorista ajureista, kuten maankäytöstä ja saasteista, 50–70 prosenttia kohdistui Suomeen. Väliraportin tulokset ovat alustavia ja ne voivat täsmentyä laskentamenetelmän kehittyessä. Väliraportin lopussa kuvataan laskentamenetelmän kehitystarpeita ja hankkeen seuraavat työvaiheet. S-ryhmän luontojalanjäljen laskenta viitoittaa tietä koko Suomen yrityskentälle. Laskennassa käytetty mittari kokoaa erilaisille lajeille aiheutetun sukupuuttoriskin yhden mittayksikön alle hiilijalanjäljen tavoin ja mahdollistaa erilaisten yritysten luontojalanjäljen kansainvälisen vertailun. Laskennan pohjalta voidaan asettaa strategisia tavoitteita luontojalanjäljen pienentämiseksi ja luontopositiivisuuden saavuttamiseksi. This intermediate report represents the first preliminary results of the biodiversity footprint assessment of S Group, initiated in 2022. The biodiversity footprint of S Group’s value chain and own operations was calculated by using a method developed by the School of Resource Wisdom, JYU.Wisdom. The basics of the method are described in the report. Results are reported for groceries, consumer durables, department stores, fuels, and hotel and restaurant services from the year 2021, and for S Group’s own operations from the year 2020. Biodiversity footprint was measured as the potentially disappeared fraction of species globally. The results showed that the biodiversity footprint of S Group increases global extinction risk for 0.0027% of species in terrestrial ecosystems, 0.00037% of species in freshwater ecosystems, and 0.011% of species in marine ecosystems. The value chain contributed to 94% of the total biodiversity footprint on terrestrial ecosystems, 92% on freshwater ecosystems and over 99% on marine ecosystems. On the other hand, S Group’s own operations contributed to 6% of the total biodiversity footprint on terrestrial ecosystems, 8% on freshwater ecosystems and less than 1% on marine ecosystems. Generally, groceries and fuels caused the largest biodiversity footprints. According to the assessment model, less than 10% of the biodiversity footprint was located in Finland, i.e., more than 90% of the biodiversity footprint was located outside of Finland. On the other hand, 50–70% of the direct drivers of biodiversity loss, such as land use and pollution, were located in Finland. The results of the report are preliminary, and they may be specified when the method is developed further. The development needs of the method and the next phases of the project are described at the end of the report. The biodiversity footprint assessment of S Group leads the way for all organizations in Finland. The indicator used in the assessment combines the extinction of various species under one indicator, similar to carbon footprints, and makes it possible to compare the biodiversity footprint of different organizations internationally. The assessment shows that it is possible to set measurable, strategic goals to mitigate the biodiversity footprint and to achieve nature positive operations.

Cancer is one of the major public health problems globally which arises due to uncontrolled cellular proliferation. Tumor necrosis factor alpha is a member of the TNF/TNFR cytokine superfamily. Currently, the venom from various sources have been widely used in the treatment of cancer. The bioactive present in bee venom has been reported to have potential antimicrobial, anti-inflammatory, and anticancer activity which has drawn the attention to identify the novel inhibitor against TNF-α. Bee venom has been reported to target ovarian, breast, prostate and malignant hepatocellular carcinoma. TNF-α is involved in the maintenance and homeostasis of the immune system, inflammation, and host defense. The oncogenic protein TNF-α plays a critical role in the development of various cancers including renal, lung, liver, prostate, bladder, and breast cancer. TNF-α enhances cancer cell growth, proliferation, invasion, and metastasis, as well as tumor angiogenesis. Due to the high prevalence and mortality, TNF-α associated cancers have remained a significant health problem globally. TNF acts biologically by activating certain signaling pathways such as nuclear factor κB (NF-κB) and c-Jun N-terminal kinase (JNK). Various toxins are being studied as alternatives for cancer treatments, and bee venom and its active components are drawing attention as potential anticancer agents. The present study identifies novel anticancer peptides that target the oncoprotein against life-threatening cancer. Docking calculations indicate that anticancer peptides, namely Melittin, Phospholipase A2, Tertiapin, and Hyaluronidase bind TNF-α respectively with the lowest binding affinity. Interestingly, Mast cell degranulating (MCD) and Apamin have the highest binding affinity with TNF-α in comparison with the above four peptides. The two lead compounds namely MCD and Apamin have the highest docking score -1253.4 and -1067.8 respectively. The present study reveals that the bee venom peptides namely MCD and Apamin interact with TNF-α associated cancer for targeted therapy of cancer. These predicted anticancer peptides are valuable candidates for in vitro or in vivo peptide therapeutic drug studies against the TNF-α associated cancers.

Post-traumatic osteoarthritis in the temporomandibular joint (TMJ OA) is associated dysfunctional cellmatrix mediated signalling resulting from changes in the pericellular microenvironment after injury. Matrix metalloproteinase (MMP)-13 is a critical enzyme in biomineralisation and the progression of OA that can both degrade the extracellular matrix and modify extracellular receptors. This study focused on MMP-13 mediated changes in a transmembrane proteoglycan, Neuron Glial antigen 2 (NG2/CSPG4). NG2/CSPG4 is a receptor for type VI collagen and a known substrate for MMP-13. In healthy articular layer chondrocytes, NG2/CSPG4 is membrane bound but becomes internalised during TMJ OA. The objective of this study was to determine if MMP-13 contributed to the cleavage and internalisation of NG2/CSPG4 during mechanical loading and OA progression. Using preclinical and clinical samples, it was shown that MMP-13 was present in a spatiotemporally consistent pattern with NG2/CSPG4 internalisation during TMJ OA. In vitro, it was illustrated that inhibiting MMP-13 prevented retention of the NG2/CSPG4 ectodomain in the extracellular matrix. Inhibiting MMP-13 promoted the accumulation of membrane-associated NG2/CSPG4 but did not affect the formation of mechanical-loading dependent variant specific fragments of the ectodomain. MMP- 13 mediated cleavage of NG2/CSPG4 is necessary to initiate clathrin-mediated internalisation of the NG2/ CSPG4 intracellular domain following mechanical loading. This mechanically sensitive MMP-13-NG2/CSPG4 axis affected the expression of key mineralisation and OA genes including bone morphogenetic protein 2, and parathyroid hormone-related protein. Together, these findings implicated MMP-13 mediated cleavage of NG2/CSPG4 in the mechanical homeostasis of mandibular condylar cartilage during the progression of degenerative arthropathies such as OA.

Aortic aneurysm rupture is typically the failure of the aortic wall to withstand the tension placed upon it by a patient’s blood pressure. Along with clinical judgement and an assessment of signs and symptoms, a computed topography angiogram (CTA) is amongst the first line of diagnostic studies having 87-94% sensitivity for rupture. Therefore, while the possibility of a CTA not catching an aortic rupture is present, the chance is very low. Usually a missed or delayed diagnosis for this type of pathology is catastrophic resulting in patient death. We present the case report of a 68-year-old male with a contained aortic rupture that was initially missed on CTA. He presented to the ED with shock like symptoms after a syncopal episode at home. Initial computed topography (CT) scan revealed a pericardial effusion with concern for hemopericardium and mass effect on the right ventricle. A follow up CTA was obtained; however, it was negative for any acute process. Given patient’s continued instability, a TTE was performed which showed concern for a dissection flap. Subsequently, a TEE was performed showing an aortic dissection at the anterior aortic root as well as severe aortic insufficiency with a perforated right coronary cusp. The patient underwent emergent cardiac surgery and recovered well despite the delay in diagnosis. This case demonstrates the importance of a high index of suspicion for acute aortic dissection despite an initial negative CTA.

Background and Aim: Non-Alcoholic Fatty Liver Disease (NAFLD) is the commonest cause of chronic liver disease and is a leading cause of liver transplantation in the United States, with no approved medication to halt or reverse its progression. Recent animal-model prospective trial-suggested that drug Vedolizumab leads to improvement and reversal in the NAFLD-related metabolic derangements. Vedolizumab is an α4β7 integrin-inhibitor that is approved for use in IBD patients. Our study aims to understand Vedolizumab's impact on the course of NAFLD in inflammatory bowel disease (IBD) patients. Methods: We conducted a retrospective cohort analysis of 158 subjects with NAFLD who received Vedolizumab at Cleveland Clinic Foundation (CCF). One cohort of 79 patients with NAFLD who received Vedolizumab were matched with control group of 79 patients. Pediatric Functional Gastrointestinal Disorders (p-FGID) comprise one of the most common chronic conditions impacting global health. Conceptualized as gut-brain communication disorders, p-FGID are often exacerbated by and contribute to significant anxiety as well as functional impairment. Cognitive behavioral therapy is the gold standard psychosocial treatment for anxiety spectrum disorders. Prior research demonstrates a bidirectional link between p-FGID and anxiety symptoms, recommending CBT as the first line of treatment for both. While exposure is the potent ingredient for anxiety treatment, exposure is under-investigated in the treatment of p-FGID. The present brief systematic review synthesizes existent literature to describe the impact of CBT on p-FGID and anxiety symptoms in youth. The researchers systematically screened articles (n=23) located via Embase, Medline, and PsychINFO search. Eight studies meeting inclusion criteria were included for the final review. Three themes characterized the selected articles: (1) the impact of CBT on p-FGID symptoms, (2) the impact of CBT on health-related disability in youth with p-FGID, and (3) the impact of CBT on concurrent anxiety symptoms in youth with p-FGID. Key findings are summarized and implications for treatment are identified.
